To use the coupon, there must be an "average" of $50 per night. For example, if you found a room that was $25 on one night and $75 the next, it will take $20 off since it "averages" $50. But if you find a room that is $75 for the first night and FREE for the second night, the average is only $37.50 and you can't apply it.
I don't know the maximum number of nights you can use the coupon in a single purchase from Expedia but it's at least 5 ($50) and could be more.
